Sildenafil citrate is an active ingredient in Viagra, Renatio or under various other trade names. It was the most commonly prescribed pharmaceuticals drugs to cure sexual deficiency and pulmonary arterial hypertension. Recent black market activity, there reported food products to contain sildenafil illegally. Instead to cure erectile dysfunction (ED), sildenafil has been used for illegal sexual activity. Side effect to abuser includes hypertension, headache, visual disturbance and even death. To avoid being detected by health ministry for illegal trafficking, this ED drug was synthesized and modified structurally, however with almost give the similar effect. This new drug is known as Sildenafil analogues. In this study, seven standards of Sildenafil analogues were obtained from Malaysian Health Department and extracted with methanol for analysis. In this study, chemical characterization was obtain using Liquid Chromatography Tandem Mass Spectrometer (LC-MSMS), while the fragmentation elucidation was done by interpreting the chromatograms obtained respectively. The advantages using LC-MSMS include its high sensitivity, specificity, and the mass spectrometer measures the characteristic ions of each analyte. This system allows the measurement of two characteristic daughter ions generated from the primary ion, thus fulfilling the identification requirements for confirmatory qualitative identification. Finally, this study will develop an LC-MSMS screening method and detection of locally prepared Sildenafil analogues.
