Abstract:

Vitamin B9 is commonly known as folic acid which is effective on skin and body system. However, there are some negative factors of folic acid due to pH sensitivity and low solubility.
Folylpolyglutamate(B9-VitapolTM) is produced to improve the negative characteristics of folic acid. Folylpolyglutamate is reformulated by combining folic acid and polyglutamate through fermentation technology. To give mass production, we have studied liquid fermentation process instead of traditional soybean fermentation process. Liquid fermentation gives high yield, less odor and less residues.
We have inoculated Bacillus subtilis from food to the medium that contains soybeans and cultured at 37 degrees celscius. We have installed the upper and lower air inlets and spargers in the fermenter.
During the first 8 hours of cultivation, operate the lower inlet and sparger to stimulate the growth of microorganism. Folic acid is added after 8 hours into the process. The lower inlet closed and the upper inlet is opened to let 2L/min of air pressure into the fermenter. Terminate stirring process and begin the static culture state for the following the 72 hours. Produced polymer is composed of folypolyglutamate and polysaccharide. Especially 50~70{6e6090cdd558c53a8bc18225ef4499fead9160abd3419ad4f137e902b483c465} of composition is folypolyglutamate. After all the processes have done, we have observed the presence of folylpolyglutamate(M.W. 200,000~1,000,000) by MALDI-TOF MS.
