Abstract: The ligninolytic enzyme activities during polycyclic aromatic hydrocarbons (PAHs) biodegradation were investigated. In the absence of PAHs Phellinussp able to shown higher ligninolytic enzyme activity only at 10th day, it decreases at 20th and 30th days. In individual PAHs degradation studies the Lignin Peroxidase (LiP); manganese peroxidase (MnP) and laccase (Lac) activity were higher at 20th day. In mixture of PAHs the maximum enzyme activity of MnPwas18.6 x 10-6 IU/ml followed by LiP (10.8 x 10-6 IU/ml) and laccase (9.6 x 10-6 IU/ml). In overall MnP was observed to be the predominant enzyme followed by LiP and Lac. The ligninolytic enzyme was purified and protein profile were analysed by using native PAGE method. In addition to that, docking studies was performed using five PAHs with LiP, MnP and Lac. These docking studies revealed that pyrene compound has high docking scores of -6.927 kcal/mol and -2.27 kcal/ mol in LiP and Laccase protein whereas MnP protein shows high docking score of -4.25 kcal/ mol with Anthracene compound. Moreover, all the docked compounds forms van der waals interactions with protein molecules hence we have reported that van der waals interactions could pave the major role in the reducing activity of three proteins.
