Abstract: In vitro propagation of Brastepu citrus with shoot tip subculture for bioconservation of local variety of citrus in North Sumatera is explained. The aim of the research is to propagate Brastepu citrus with shoot tip subculture to obtain citrus seedling as a strategy in the bioconservation of local variety of citrus in North Sumatra. The research is conducted experimentally with variation of subculture frequency. It was found that the best condition to obtain callus was obtained by using combination of 0.5 mg/L 2,4-D with 0.5 mg/L BAP. Embryosomatic was obtained when the treatment combination is performed at 1.0 mg/L 2,4-D with 0.5 mg/L BAP. The best condition of shoot was produced when 0.5 mg/L 2,4-D and 1.5 mg/L BAP are applied. In vitro propagation with shoot tip subculture for one time is found the best condition to produce good quality seedling. The variation in treatment conditions were significantly influenced the growth and development of callus, embryosomatic, and the shoot of Brastepu citrus.
