Abstract:
Ondensetron(OND) is a weakly basic (BCS I) drug used in treatment of irritable bowel syndrome (IBS).Currently used colon targeted marketed formulation suffer from drawbacks like variable transit time, absence of enzymes and occasional drop in pH. In this project we have prepared mucoadhesive multiparticulate
System (MPS) of ondansetron with dual sensitive coating combining both pH triggered (provided by Eudragit S100) and bacterial enzyme triggered (due to presence of gaur gum) system to provide drug release at intended site of action.
Pellets of OND were prepared using spheronizer -extruder and were evaluated for parameters like useable yield, abrasion resistance, residual moisture, in-vitro drug release and ex-vivo mucoadhesion. Surface morphology characterization of the developed pellets was also done using SEM.
The results revealed 60{6e6090cdd558c53a8bc18225ef4499fead9160abd3419ad4f137e902b483c465} usable yield of the pellets with less than 1{6e6090cdd558c53a8bc18225ef4499fead9160abd3419ad4f137e902b483c465} abrasion. Core Pellets having 15 {6e6090cdd558c53a8bc18225ef4499fead9160abd3419ad4f137e902b483c465} of Carbopol 974 P exhibited good mucoadhesion behavior, and the mucoadhesion tends to decrease with increase in concentration of Carbopol 974 P. In-vitro dissolution revealed that 25{6e6090cdd558c53a8bc18225ef4499fead9160abd3419ad4f137e902b483c465} w/w of Gaur gum and Eudragit S100 was required to achieve the targeted profile of delayed release of OND in colon.SEM images revealed smooth and uniform coating on pellets.
