Abstract: We investigated the synaptic plasticity effects of FK506 in organotypic hippocampal slice culture (OHSC) using optical imaging system. Hippocampal slices were prepared from 6-8 day rats using a tissue chopper and placed on a membrane insert. After 18 hr KA (kainic acid) treatment, significant delayed neuronal death which was quantified by PI (propidium iodide) staining was detected in CA3 and CA1 region at 24 hr after KA treatment. The neuronal death was significantly prevented at 24 hr after 0.1 μM FK506 treatment. In electrophysiology study, the optical signals following long-term potentiation (LTP) was observed by Schaffer collateral pathway stimulation. The improvement in amount of LTP was found in FK506-treated group. These results suggest that FK506 may have a positive role in synaptic efficacy following KA-induced neuronal cell death.
