Abstract: In this project immobilized thermostable -glucosidases were used from Thermotoga neapolitana (Bgl1A of Glycoside hydrolase family 1 (GH1)) to catalyze the hydrolysis of polyphenolic antioxidative molecules. In order to explore the possibilities to utilize these thermostable beta-glucosidases in immobilized form and to design an online mini flow beta-glucosidase system for deglycosylation of quercetin, two different approaches to covalent immobilization of enzyme have been explored: (a) a direct enzyme binding to polymers via their oxirane groups (b) enzyme binding to polymers via spacer made from ethylene diamine/ glutaraldehyde. The three epoxy-activated acrylic particulate polymers used in the study were Eupergit C, Eupergit C 250L and cryogels. The performances of these immobilized catalysts were compared with respect to specific activity (U/mg) at different temperature 80-95C.
