Abstract:

To establish a superovulation procedure for in golden hamster, harvesting high quantity of oocytes in perfect stage of maturation under uncontrolled condition, Hamsters were assigned to eight groups and superovulated with pregnant mare serum gonadotropin (PMSG; 40 IU), without regards to the estrous cycle and (48-52 h or 52-72 h) later on injected with human chorionic gonadotropin (hCG; 40 or 45 IU), (12-15 h, or 16-18 h) after second injection, anesthesia and oocyte recovery. Significant statistical differences (p > 0.05) were found between different programs. In program eight, the number of harvested oocytes (66 1.2) were significantly higher (P 0.05) was found on fertilization percentage (FP) and fertilization index (FI) in
all programs. From these results, we concluded that administration of 40 IU PMSG on Day 1 experiment (without regard to estrous cycle), 52-72 h later hCG, and anesthesia 16-18 h, for superovulated hamsters in tropical climate is applicable. Number of oocyte recovery was increased with no adverse effect on oocyte quality
