Abstract:

The effect of lyophilization on the asymmetric reduction reaction catalyzed by immobilized carrots was investigated. In this work, we studied asymmetric reduction of prochiral aromatic ketone to produce (S)-1-phenylethanol with immobilized freeze-dried carrot as the biocatalyst. Acetophenone (ACP) was chosen as the model substrate. The results show that acetophenone can be reduced to the chiral alcohols with high enantioselectivity by the immobilized freeze-dried carrot. In the reaction, (S)-1-phenylethanol is produced with immobilized freeze-dried carrot.
Also, the reaction characteristics were studied in detail using immobilized freeze-dried carrot as the biocatalyst. The effects of various factors on the reaction were investigated such as initial substrate concentration, reaction time, temperature and pH. Best results for enantiomeric excess (ee) and conversion (c) were obtained >99{6e6090cdd558c53a8bc18225ef4499fead9160abd3419ad4f137e902b483c465} and 50-62{6e6090cdd558c53a8bc18225ef4499fead9160abd3419ad4f137e902b483c465} as reaction time 48 h, initial substrate concentration 0.5 mmol·L-1, reaction temperature 33 oC and pH 7.
In the same study, cofactor requirement was investigated to lyophilized carrots. Glucose dehydrogenase (GDH) enzyme and glucose molecule in plant cells can provide cofactor regeneration.
