Abstract: Sildenafil citrate, one of the phosphodiesterase-5 inhibitor, is considered the effective medicine for erectile dysfunction it has been reported that its pharmacokinetics are affected by hepatic impairment. But the role of sildenafil on the liver architecture is not much explored. Therefore the present study was undertaken to study the structure of liver after administration of sildenafil in wistar rats. The animals were divided into a control group and four experimental group containing six rats in each group. The animals were treated with sildenafil citrate dissolved in distilled water. The microscopic structure and certain biochemical parameters in kidney were studied. The results show that at low dosage the hepatic architecture is not much altered were as as the dosage and duration of exposure of sildenafil increased the liver showed structural alterations which was linked to change in biochemical parameters.
