Abstract:
Design a kind of protein microarrays for parallel detection of multiple viral antigens and antibodies of veterinary medicines. Determined the best probe fixed concentration and pH value and study the evaluation indexes of this method, including sensitivity, specific degree, repeatability and other basic parameters of SPR biological sensors testing system. The optimization immobilization pH value and concentration were 5.0 and 0.5mg/ml respectively. There was no cross-reaction between antibodies; coefficient of variation(CV) interarrays and intraarrays were 2.87{6e6090cdd558c53a8bc18225ef4499fead9160abd3419ad4f137e902b483c465} and 3.84{6e6090cdd558c53a8bc18225ef4499fead9160abd3419ad4f137e902b483c465} respectively; the detection sensitivity can reach 0.1ng/ml. the SPR sensor’s sensitivity and specificity were 96.4{6e6090cdd558c53a8bc18225ef4499fead9160abd3419ad4f137e902b483c465}(27/28)and 90.9{6e6090cdd558c53a8bc18225ef4499fead9160abd3419ad4f137e902b483c465}(30/33) respectively.
