The main purpose of this study was to determine the heterogeneity of primary isolated mononucleated cells using molecular markers approach. This study was performed using mononucleated cells originated from murine central blood system. The isolated cells were cultured in complete medium for 14 days. During the culturing days, the cells were transferred into a new well-plate for about 4 to 7 days prior to separate different forms of cells i.e., adherent and suspension forms. Both hematopoietic and mesenchymal stem cells molecular markers activation were examined on cells after 14 days cultured in
complete medium using RT-PCR reaction. After being separated and cultured for 14 days, we found that the adherent cells activate the Cd105 and Cd73 while suspension cells showed the activation of Cd38 and Sca-1 genes. As a conclusion, molecular analysis showed that isolated mononucleated cells are consist of hematopoitic stem cells (suspension) and mesenchymal stem cells (adherent).
